Among these techniques, Sanger sequencing (also known as chain-termination sequencing) stands as a landmark achievement that laid the foundation for unraveling genetic mysteries.

The Nobel Prize-Winning Breakthrough

Developed in 1977 by British biochemist Frederick Sanger and his colleagues, this revolutionary method earned Sanger his second Nobel Prize in Chemistry. The technique's brilliance lies in its elegant simplicity: using DNA polymerase to synthesize new DNA strands while incorporating special terminating nucleotides (ddNTPs).

These ddNTPs lack a 3'-hydroxyl group, causing immediate chain termination when incorporated into growing DNA strands. By including four distinct ddNTPs (ddATP, ddTTP, ddCTP, and ddGTP)—each labeled with different fluorescent dyes or radioactive isotopes—researchers generate DNA fragments of varying lengths, each ending with a specific ddNTP.

From Gel Electrophoresis to Modern Automation

The DNA fragments are then separated by size through gel electrophoresis, with shorter fragments migrating faster. By detecting the terminal markers of these fragments, scientists can reconstruct the original DNA template's base sequence. Early implementations relied on radioactive isotope labeling and gel imaging—a laborious process. Technological advancements later introduced fluorescent labeling and laser scanning, enabling automation that significantly improved efficiency and accuracy.

Modern Sanger sequencers can process multiple samples simultaneously, rapidly generating electropherograms (commonly called "peak traces") that visually represent DNA sequences.

Why Sanger Sequencing Still Matters

Despite being surpassed in throughput by high-throughput sequencing technologies (such as next-generation sequencing), Sanger sequencing maintains critical advantages that ensure its continued relevance. With superior accuracy, relatively long read lengths (500-1000 base pairs), straightforward protocols, and cost-effectiveness for small-scale projects, it remains indispensable for numerous applications.

The technique serves as the gold standard for gene cloning verification, mutation detection, genotyping, PCR product validation, and small-scale genome studies. Its contributions continue to support genetic research, medical diagnostics, and advancements in biotechnology.
